Grave of the Fireflies
Hands down, without a doubt -- the saddest movie of all time. This is a one timer, and not just because it's sad. Because the images will stay with you.

Powerful, tragic, human, and a bold anti-war statement, Grave of the Fireflies tells the story of two orphaned siblings trying to survive in Japan at the end of World War II. I have never cried more at a movie than I did at this. It's been several years since I've seen it, and though at the time I said "NEVER AGAIN!", I've been thinking of watching it again. You know, when I have a full day to do nothing but sit on my couch and sob openly, curled in the fetal position.
In all, a profoundly affecting movie that everyone should see once. You'll be surprised at how human everyone seems, even through the anime.
